چکیده

6-Methylthiopurine ribonucleoside was found to inhibit nucleotide formation from hypoxanthine and aminoimidazole carboxamide in Ehrlich ascites tumor cells in vitro and to inhibit inosinate dehydrogenase activity in intact cells. These effects are produced at higher drug concentrations than required to inhibit purine biosynthesis de novo. 4-Methylthio-7-|8-D-ribofuranosyl pyrrolo[2,3-i/]pyrimidine also inhibited nucleotide formation and inosinate dehydrogenase activity, and several other methylthio purine ribonucleoside analogs or derivatives also inhibited one or another of these processes.

Inhibition of Furine Ribonucleotide and Phosphoribosyl Pyrophosphate Synthesis by 6-CycIopentylthio-9- hydroxymethylpurine and Structurally Related Compounds1

o-Cyclopentylthio-Q-hydroxymethylpurine, 9-(3-aminopropyl)adenine, and structurally related compounds in hibited the conversion of adenine, guanine, and hypoxanthine to ribonucleotides in Ehrlich ascites tumor cells in vitro. These compounds also inhibited the accumulation of phosphoribosyl pyrophosphate, a substrate required for ribonucleotide formation from purines. 6-Cyclopentylthio9-hydroxy...

Reversal of methylmercaptopurine ribonucleoside cytotoxicity by purine ribonucleosides and adenine.

6-Methylmercaptopurine ribonucleoside-5'-phosphate (MeSPuRMP), the sole metabolite of 6-methylmercaptopurine ribonucleoside (MeSPuRib), is a strong inhibitor of purine de novo synthesis, inducing depletion of intracellular purine nucleotides and subsequent cell death in several tumor cell lines. In this study prevention of MeSPuRib cytotoxicity by compounds of the purine salvage pathway was stu...

Synergistic Effect of 6-Mercaptopurine and 6-Methylmercaptopurine Ribonucleoside on the Levels of Adenine Nucleotides of Sarcoma 180 Cells1

The synergistic antitumor action of 6-mercaptopurine (6-MP) and 6-methylmercaptopurine ribonucleoside (MMPR) is confirmed with Sarcoma 180 ascites cells. Striking decreases are seen in the adenine nucleotide concentrations in cells incubated for 1 hr with this drug combination, and lesser effects are seen on the guanine nucleotide concentrations. Still greater decreases in adenine nucleotide le...

Furine Nucleotide Metabolism and Nucleotide Pool Sizes in Synchronized Lymphoma L5178Y Cells1

Mouse lymphoma L5178Y cells in culture were syn chronized by exposure to thymidine and Colcemid. At four times during the cell cycle, various aspects of purine metab olism were examined with the use of radioactive purine bases. In all experiments the uptake and conversion of bases to nucleotides was rate limiting for ribonucleoside triphosphate synthesis. Cell cycle variations in the apparent e...
